Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of MLL peptide (Run 1) or non-specific control peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1061, Anti-MLL/HRX CT, clone 5C15 ZooMAb® Rabbit Monoclonal. The binding of this antibody to MLL peptide displayed a KD of 1.2 x 10-6, whereas binding to non-specific control peptide displayed a KD of 6.7 x 10-3.

Target description: Histone-lysine N-methyltransferase 2A (UniProt: Q03164; also known as EC:2.1.1.354, Lysine N-methyltransferase 2A, ALL-1, CXXC-type zinc finger protein 7, Myeloid/lymphoid or mixed-lineage leukemia, Myeloid/lymphoid or mixed-lineage leukemia protein 1, MLL, Trithorax-like protein, Zinc finger protein HRX) is encoded by the KMT2A (also known as ALL1, CXXC7, HRX, HTRX, MLL, MLL1, TRX1) gene (Gene ID: 4297) in human. MLL is a histone methyltransferase that plays an essential role in early development and hematopoiesis. It is expressed in heart, lung, brain, and in T and B lymphocytes. Catalytic subunit of the MLL complex mediates both methylation of lysine 4 of histone H3 (H3K4me) complex and acetylation of lysine 16 of histone H4 (H4K16ac). By itself it has weak methyltransferase activity and requires another component of the MLL complex to obtain full methyltransferase activity. MLL does not display any activity towards histone H3 phosphorylated on threonine 3 and has less activity towards histone H3 dimethylated on arginine 8 and lysine 9. However, it displays high activity towards histone H3 acetylated on lysine 9. MLL is normally cleaved into a N-terminal p320 and a C-terminal p180 cleavage product. This cleavage is brought about by taspase 1 (TASP1) at cleavage site 1 (aa 2666-2667) and cleavage site 2 (aa 2718-2719). The p320 and p180 units can heterodimerize via SET and FYRC domains.
