Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from HeLa (Lane 1), HepG2 (Lane 2), and COS-7 (Lane 3) cells were probed with Cat. No. ZRB1052, Anti-FTO, clone 4B21 ZooMAb® Rabbit Monoclonal (1:10,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates FTO (~53 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 μg/mL of non-specific control peptide (Run 1) or FTO peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1052, Anti-FTO, clone 4B21 ZooMAb® Rabbit Monoclonal. The binding of this antibody to FTO peptide displayed a KD value of 4.1 x 10-8 whereas its binding to non-specific control peptide displayed a KD of 2.9 x 10-4.

Target description: Alpha-ketoglutarate-dependent dioxygenase FTO (UniProt: Q9C0B1; also known as Fat mass and obesity-associated protein, U6 small nuclear RNA (2′-O-methyladenosine-N(6)-)-demethylase FTO, U6 small nuclear RNA N(6)-methyladenosine-demethylase FTO, mRNA (2′-O-methyladenosine-N(6)-)-demethylase FTO, m6A(m)-demethylase FTO, mRNA N(6)-methyladenosine demethylase FTO, tRNA N1-methyl adenine demethylase FTO) is encoded by the FTO (also known as KIAA1752) gene (Gene ID: 79068) in human. FTO is a ubiquitously expressed protein, but relatively higher expression is observed in adrenal glands and brain; especially in hypothalamus and pituitary. It is mainly localized in the nucleus where it serves as an RNA demethylase and mediates oxidative demethylation of different RNA species and acts as a regulator of fat mass, adipogenesis and energy homeostasis. It is shown to specifically demethylate N6-methyladenosine (m6A) RNA, which is one of the most prevalent internal modification of messenger RNA in higher eukaryotes. FTO can also act as a tRNA demethylase by removing N1-methyladenine from various tRNAs. However, it does not display any activity towards 1-methylguanine and double-stranded DNA. FTO is involved in the regulation of thermogenesis, fat mass, adipogenesis and body weight, thereby contributes to the regulation of body size and body fat accumulation. It is also reported to control adipocyte differentiation into brown or white fat cells. Its activity is enhanced in the presence of ascorbate and inhibited by N-oxalylglycine, fumarate and succinate. Mutations in FTO gene or altered expression of adjacent genes, such as IRX3, have been linked to obesity. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
