Target description: | Phospholysine phosphohistidine inorganic pyrophosphate phosphatase (UniProt: Q9H008; also known as EC:3.6.1.1, Hlhpp, LHPP) is encoded by the LHPP gene (Gene ID: 64077) in human. LHPP is an evolutionarily conserved member of the HAD-like hydrolase superfamily with phosphatase activity. It displays broad substrate specificity and can hydrolyze imidodiphosphate, 3-phosphohistidine, and 6-phospholysine. It can also hydrolyze inorganic diphosphate, but with reduced efficiency. It has two substrate binding sites that are located in amino acids 17-19 and 54-55. Its higher expression has been reported in brain, and somewhat lower expression in liver and kidney. LHHP expression is shown to be down regulated in liver and bladder cancers and knockdown of LHPP promotes the proliferation and growth of tumor cells. Conversely, sustained hepatic expression of LHPP in a murine model of hepatocellular carcinoma is reported to reduce tumor burden and prevent the loss of liver function. Silencing of LHHP can lead to enhanced phosphorylation of Akt and p65 and up-regulates the level of Bcl-2 and cyclin D1. Two isoforms of LHPP have been described that are produced by alternative splicing.
