Immunohistochemistry (Paraffin) Enhanced Validation-Recombinant Antibody Technology Formalin Fixed Paraffin Embedded (FFPE) human liver (A) and Negative control (no primary) (B) tissue sections were prepared using heat-induced epitope retrieval (HIER). Immunostaining was performed using a 1:1,000 dilution of Cat. No. ZRB1050, Anti-LHPP, clone 3B18 ZooMAb® Rabbit Monoclonal. Reactivity was detected using an Goat Anti-Rabbit IgG and HRP-DAB. Cytoplasmic/nuclear staining was observed in hepatocytes of human liver tissue sections.

Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from A431 cells (Lane 1), Rat kidney (Lane 2), Human liver (Lane 3), and COS-7 cells (Lane 4) were probed with Cat. No. ZRB1050, Anti-LHPP, clone 3B18 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates LHPP (~30 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 μg/mL of recombinant human LHPP protein (Run 1) and non-specific GST protein (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1050, Anti-LHPP, clone 3B18 ZooMAb® Rabbit Monoclonal. The binding of this antibody to recombinant LHPP displayed a KD value of 8.6 x 107 whereas its binding to non-specific GST protein displayed a KD of 7.6 x 104.

Target description: Phospholysine phosphohistidine inorganic pyrophosphate phosphatase (UniProt: Q9H008; also known as EC:3.6.1.1, Hlhpp, LHPP) is encoded by the LHPP gene (Gene ID: 64077) in human. LHPP is an evolutionarily conserved member of the HAD-like hydrolase superfamily with phosphatase activity. It displays broad substrate specificity and can hydrolyze imidodiphosphate, 3-phosphohistidine, and 6-phospholysine. It can also hydrolyze inorganic diphosphate, but with reduced efficiency. It has two substrate binding sites that are located in amino acids 17-19 and 54-55. Its higher expression has been reported in brain, and somewhat lower expression in liver and kidney. LHHP expression is shown to be down regulated in liver and bladder cancers and knockdown of LHPP promotes the proliferation and growth of tumor cells. Conversely, sustained hepatic expression of LHPP in a murine model of hepatocellular carcinoma is reported to reduce tumor burden and prevent the loss of liver function. Silencing of LHHP can lead to enhanced phosphorylation of Akt and p65 and up-regulates the level of Bcl-2 and cyclin D1. Two isoforms of LHPP have been described that are produced by alternative splicing. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
