Target description: | Sialic acid-binding Ig-like lectin 9 (UniProt: Q9Y336; also known as Siglec-9, CDw329, Protein FOAP-9, CD329) is encoded by the SIGLEC9 (also known as UNQ668/PRO1302) gene (Gene ID: 27180) in human. Sialic-acid-binding immunoglobulin-like lectins (Siglecs) belong to a lectin family of surface receptors that are predominantly expressed on hematopoietic cells with cell type and differentiation specificity. By binding with sialic acids at the terminal position of glycoproteins and lipids they regulate the intensity and duration of innate or adaptive immune response. Siglec-9 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-17), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 18-348), a transmembrane domain (aa 349-369), and a cytoplasmic domain (aa 370-463). It has one Ig-like V-type domain (aa 20-140) and two Ig-like C2-type domains (aa 146-229 and 236-336). It also contains one ITIM motif (aa 431-436) that is involved in modulation of cellular responses. The phosphorylated ITIM motif can bind the SH2 domain of several SH2-continaing phosphatases. It is expressed by peripheral blood leukocytes and is also found in a variety of other tissues at lower levels. Siglec9 serves as a putative adhesion molecule that mediate sialic-acid dependent binding to cells. It preferentially binds to alpha-2,3- or alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. Reduced expression of Siglec-9 on natural killer cells has been linked to persistent hepatitis B virus replication. Increased expression of soluble Siglec-9 has been reported in subjects with chronic obstructive pulmonary disease.
