Target description: | Sialic acid-binding Ig-like lectin 7 (UniProt: Q9Y286; also known as Siglec-7 Adhesion inhibitory receptor molecule 1, AIRM-1, CDw328, D-siglec, QA79 membrane protein, p75, CD328) is encoded by the SIGLEC7 (also known as AIRM1) gene (Gene ID: 27036) in human. Siglec-7 is a single-pass type I membrane glycoprotein protein that is predominantly expressed by resting and activated natural killer (NK) cells. Its high expression is also observed in placenta, liver, lung, spleen, and peripheral blood leukocytes. Siglec-7 serves as an inhibitory receptor expressed on NK cells. Siglec-7 is also expressed on β-cells where it is shown to restore β-cell function and survival and reduce inflammation in pancreatic islets from patients with diabetes. Its levels are down-regulated in Type 1 and Type 2 diabetes. It preferentially binds to alpha-2,3- and alpha-2,6-linked sialic acid and also binds disialogangliosides. Siglec-7 is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to generate the mature form that has an extracellular domain (aa 19-353), a transmembrane domain (aa 354-376), and a cytoplasmic domain (aa 377-467). Siglec-7 also contains one copy of immunoreceptor tyrosine-based inhibitor motif (ITIM) that is involved in modulation its cellular responses.
