Cytoplasmic/membranous staining in both germinal center and non-germinal center cells of human tonsil tissue sections.

No. ZRB1011, Anti-CD3e, clone 1H4 ZooMAb® Rabbit Monoclonal and visualized with a secondary antibody conjugated to Alexa Fluor® 488 (Green). Nucleus is stained with DAPI (Blue). This antibody positively stains the plasma membrane.
Western Blotting Enhanced Validation-Recombinant Antibody Technology Jurkat cell lysate was probed with Cat. No. ZRB1011, Anti-CD3e, clone 1H4 ZooMAb® Rabbit Monoclonal (1:2,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates CD3e (~21 kDa).
Flow Cytometry Enhanced Validation-Recombinant Antibody Technology Staining of one million Jurkat cell line was performed using 0.01 μg of Cat. No. ZRB1011, Anti-CD3e, clone 1H4 ZooMAb® Rabbit Monoclonal (Yellow histogram) or the equivalent amount of a Rabbit IgG isotype control (Grey histogram), followed by a PE-conjugated Donkey Anti-Rabbit secondary antibody.

Target description: T-cell surface glycoprotein CD3 epsilon chain (UniProt P07766; also known as CD3-epsilon, CD3e, T-cell surface antigen T3/Leu-4 epsilon chain) is encoded by the CD3E (also known as T3E) gene (Gene ID 916) in human. The T cell receptor or TCR is a plasma membrane protein complex responsible for recognizing antigens bound to major histocompatibility complex (MHC) on antigen presenting cells. In 95% of T cells, the TCR is composed of a disulfide-linked alpha and beta dimer, while among 5% of T cells the TCR is composed of a gamma and a delta dimer. T cells are activated when their surface TCRs engage with antigen/MHC complex on antigen presenting cells. Each TCR alpha and beta chain contains a variable (V) and a constant (C) extracellular immunoglobulin-like domain, followed by a transmembrane region and a short cytoplasmic tail. The alpha and beta chains have very short cytoplasmic tail and rely on the CD3 (cluster of differentiation 3) co-receptor complex for transducing signaling further intracellularly. The CD3 co-receptor complex is composed of a CD3 chain, a CD3 chain, and two CD3 chains. The CD3 complex, TCR dimer, and a -chain together comprise the functional TCR complex. The CD3 epsilon chain is initially produced with a signal peptide (aa 1-22), the removal of which yields the mature chain (aa. 23-207) with an Ig-like (aa 32-112) domain-containing extracellular region (aa 23-126), a transmembrane segment (aa 127-152), and a cytoplasmic tail (aa 153-207), which contains the immunoreceptor tyrosine-based activation motif or ITAM (a.a. 178-205) that is essential for mediating ZAP-70 interaction and transducing TCR signaling. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
