Cytoplasmic/membranous staining was observed in non-germinal center cells of human tonsil tissue sections.

Western Blotting Enhanced Validation-Recombinant Antibody Technology Human tonsil (Lane 1) and THP-1 cell (Lane 2) lysates were probed with Cat. No. ZRB1010, Anti-CD8a, clone 1B19 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates CD8a (~35 kDa).

Target description: T-cell surface glycoprotein CD8 alpha chain (UniProt: P01732; also known as T-lymphocyte differentiation antigen T8/Leu-2, CD8a) is encoded by the CD8A (also known as MAL) gene (Gene ID: 925) in human. CD8a is a single-pass type I integral membrane protein with one Ig-like V-type domain (aa 22-135) that plays an essential role in the immune response and serves multiple functions in responses against both external and internal offenses. CD8a is produced with a 21-amino acid signal peptide that is subsequently cleaved to produce the mature form. CD8 is generally present as a heterodimer of alpha and beta chains linked by two disulfide bonds, however, it can also form homodimers. The CD8 alpha/alpha homodimer is expressed on a subset of gamma/delta TCR-bearing T cells, NK cells, intestinal intraepithelial lymphocytes, and lymphoid dendritic cells. CD8a functions primarily as a co-receptor for MHC class I molecule:peptide complex. CD8a is reported to interact simultaneously with the T-cell receptor (TCR) and the MHC class I proteins presented by antigen presenting cells. In NK-cells, the presence of CD8a homodimers at the cell surface provides a survival mechanism allowing conjugation and lysis of multiple target cells. CD8a homodimer molecules also promote the survival and differentiation of activated lymphocytes into memory CD8 T-cells. Cytoplasmic domains of CD8 provide binding sites for the tyrosine kinase lck, facilitating intracellular signaling events that lead to T cell activation, development, and cytotoxic effector functions. CD8+ cytotoxic T cells play an important role in inducing cell death of tumor cells, as well as cells infected by virus, bacteria or parasites. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
