Immunohistochemistry (Paraffin) Enhanced Validation-Recombinant Antibody Technology Formalin Fixed Paraffin Embedded (FFPE) rat colon (A) and rat cerebral cortex (B) tissue sections were prepared using heat-induced epitope retrieval (HIER). Immunostaining was performed using a 1:100 dilution of Cat. No. ZRB06182, Anti-p44 MAPK (ERK1), clone 13F10 ZooMAb® Rabbit Monoclonal. Reactivity was detected using a Goat Anti-Rabbit and DAB. Cytoplasmic/membranous/nuclear staining was observed in glandular cells of rat colon and neurons of rat cerebral cortex tissue sections.

Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from Rat L6 (Lane 1), Rat C6 (Lane 2), and Mouse NIH/3T3 (Lane 3) cells were probed with Cat. No. ZRB06182, Anti-p44 MAPK (ERK1), clone 13F10 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates p44 MAPK (ERK1) (~44 and 42 kDa).
Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysate from Human A431 cells was probed with Cat. No. ZRB06182, Anti-p44 MAPK (ERK1), clone 13F10 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates p44 MAPK (ERK1) (~44 and 42 kDa).
Immunoprecipitation Enhanced Validation-Recombinant Antibody Technology 100 μL of Cat. No. ZRB06182, Anti-p44 MAPK (ERK1), clone 13F10 ZooMAb® Rabbit Monoclonal was used to immunoprecipitate p44 MAPK (ERK1) in 500 μg of Rat L6 cell lysate. Immunoprecipitation reaction using L6 cell lysate and ZooMAb® Anti-p44-42 MAPK. Image is 30 sec exposure using forte. Lane 1 is 2% of input (10 μg of L6 lysate). Lane 2 is 15% of IP reaction (15 μL out of 100 run in lane) were then probed with Cat. No. ZRB06182, Anti-p44 MAPK (ERK1), clone 13F10 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using an Anti-Rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates p44 MAPK (ERK1) (~44 and 42 kDa).

Target description: Mitogen-activated protein kinase 3 (UniProt: P21708; also known as EC: 2.7.11.24, MAP kinase 3, MAPK 3, ERT2, Extracellular signal-regulated kinase 1, ERK-1, Insulin-stimulated MAP2 kinase, MAP kinase isoform p44, p44-MAPK, MNK1, Microtubule-associated protein 2 kinase, p44-ERK1) is encoded by the Mapk3 (also known as Erk1, Prkm3) gene (Gene ID: 50689) in rat. ERK-1 is a serine/threonine kinase that acts as an essential component of the MAP kinase signal transduction pathway. It is expressed in different tissues, mostly in intestine, placenta, and lung. Highest levels have been reported in the nervous system. The MAPK/ERK cascade mediates several diverse biological processes such as cell growth, adhesion, survival, and differentiation through the regulation of transcription, translation, and cytoskeletal rearrangements. It also plays also a role in initiation and regulation of meiosis, mitosis, and post-mitotic functions in differentiated cells by phosphorylating a few transcription factors. About 160 substrates have been described for ERKs. The protein kinase domain of ERK-1 is localized to amino acids 43-331 and the TXY motif is located in amino acids 203-205. ERK-1 is phosphorylated by MEK1 and MEK2 on threonine 203 and tyrosine 205 in response to external stimuli like insulin or nerve growth factor and both these phosphorylations are essential for its activity. Two isoforms of ERK-1 have been described that are produced by alternative splicing.
