Immunohistochemistry (Paraffin) Enhanced Validation-Recombinant Antibody Technology Formalin Fixed Paraffin Embedded (FFPE) human pancreas tissue sections were prepared using heat-induced epitope retrieval (HIER). Immunostaining was performed using a 1:100 dilution of Cat. No. ZRB05665, Anti-beta Catenin, clone 3J7 ZooMAb® Rabbit Monoclonal. Reactivity was detected using a Goat Anti-Rabbit and DAB. Cytoplasmic/membranous staining was observed in both acinar & islet cells of human pancreas tissue sections.
Immunocytochemistry Enhanced Validation-Recombinant Antibody Technology Immunofluorescent analysis of A431 cells was performed using a 1:100 dilution of Cat. No. ZRB05665, Anti-beta Catenin, clone 3J7 ZooMAb® Rabbit Monoclonal and visualized with a Goat Anti-Rabbit secondary antibody conjugated to Alexa Fluor® 488 (Green). Actin filaments have been labeled with phalloidin (Red). Nucleus is stained with DAPI (Blue). This antibody positively stains the cell membrane.

Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from HeLa cells (A) and NIH3T3 cells (B) were probed with Cat. No. ZRB05665, Anti-beta Catenin, clone 3J7 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates beta Catenin (~85 kDa).

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Specificity: Clone 3J7 is a ZooMAb rabbit recombinant monoclonal antibody that detects beta-catenin. It targets an epitope with in the N-terminal region of isoform 1.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Catenin beta-1 (UniProt: P35222; also known as beta-catenin) is encoded by the CTNNB1 (also known as CTNNB, OK/SW-cl.35, PRO2286) gene (Gene ID: 1499) in human. Catenin beta-1 is a key downstream component of the canonical Wnt signaling pathway that participates in intercellular adhesion and Wnt-mediated transcriptional activation. In the absence of Wnt, it forms a complex with AXIN1, AXIN2, APC, CSNK1A1, and GSK-3 beta that promotes phosphorylation on N-terminal serine and threonine residues that leads to its subsequent degradation by the proteasome. In the presence of Wnt ligand, it is not ubiquitinated and accumulates in the nucleus, where it acts as a coactivator for transcription factors of the TCF/LEF family, which leads to activation of Wnt responsive genes. O-glycosylation at serine 23 is reported to decrease its nuclear localization and transcriptional activity. Wnt/beta-catenin signaling is shown to be essential for embryonic stem cell (ESC) pluripotency and differentiation. In ESC beta-catenin is trimethylated on Lysine 49 by Ezh2 methyltransferase and this trimethylated form represses neuronal differentiation genes Sox1 and Sox3. Mutations in CTNNB1 gene are shown to cause colorectal cancers, medulloblastoma, and ovarian cancers. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
