Immunofluorescence localization of Dicer in HeLa cells. Cells were stained with 5 μg/mL Rabbit Anti-Dicer (Cat. No. SAB4200087) followed by Goat Anti-Rabbit, Cye3 conjugate. The cells were counterstained with DAPI (blue) to stain nuclei (×40 magnification).
Immunoblotting HeLa cells lysate was separated on SDS-PAGE and probed with Rabbit Anti-Dicer (Cat. No. SAB4200087). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescence substrate. Lanes 1. Antibody 2 μg/mL 2. Antibody 4 μg/mL 3. Antibody 4 μg/mL + 20 μg/mL Dicer immunizing peptide
Immunoprecipitation Rabbit Anti-Dicer (Cat. No. SAB4200087) was used to immunoprecipitate human Dicer from HeLa cells. Lanes 1. IP antibody: 2.5 μg 2. IP antibody: 5 μg 3. Negative control: IP antibody without cell lysate 4. Negative control: without IP antibody

Biochem/physiol Actions: Anti-Dicer recognizes human Dicer.
Biochem/physiol Actions: Dicer catalyzes the first step in the RNA interference (RNAi) pathway and initiates formation of the RNA-induced silencing complex (RISC). Dicer processes the dsRNA into small fragments called short interfering RNA (siRNA) or microRNA (miRNA) of typically 21-25 nucleotides long with a two-base overhang on the 3′. It helps to load these fragments into the RNA-induced silencing complex (RISC), which guides the sequence-specific silencing of mRNAs that contain complementary sequences by either enzymatically cleaving the mRNA or repressing its translation.
Biochem/physiol Actions: Mutation in the Dicer gene causes malignancies.

General description: Dicer, also known as Dicer1, endoribonuclease Dicer, Helicase with RNase motif is a member of the RNase III family. It contains a N-terminal DEXH-box RNA helicase/ ATPase domain, followed by a domain of unknown function (DUF283), a Piwi/Argonaut/Zwill (PAZ ) domain, which anchors the 3′-end of RNA substrates.
General description: Dicer1 is encoded by the gene mapped to human chromosome 14q32.13.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note: Store at –20 °C. For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
