Immunoblotting
Whole cell extracts from staurosporine-treated NIH3T3 (Lane A) and HeLa (Lanes B, C, D) cells were separated on SDS-PAGE, blotted with Rabbit Anti-phospho-Histone H2AX [pSer139] (Cat. No. H5912) and developed using Goat Anti-Rabbit IgG Peroxidase (Cat. No. A0545) and chemiluminescent substrate. The antibody is specific for the pSer139 phosphorylated Histone H2AX as shown by the inhibition of antibody binding with the phosphorylated peptide and lack of inhibition with the non-phosphorylated peptide. (Lane A,C) Antibody dilution 1:1,000; (Lane C) Antibody dilution 1:1,000+ 5 μg/ml phospho-Histone H2AX [pSer139] peptide; (Lane D Antibody dilution 1:1,000+ 5 μg/ml non-phospho-Histone H2AX peptide.

Biochem/physiol Actions: H2AX is critical for facilitating the assembly of specific DNA-repair complexes on damaged DNA. Ataxia-telangiectasia-mutated (ATM) is the major kinase involved in the phosphorylation of H2AX in response to DNA double-strand breaks. H2AX knockout mice are radiation sensitive and show retarted growth. They also are immune deficient with mutant males are infertile..

General description: H2AX comprises 2-25% of the total H2A complement in human cells, whereas in the budding yeast it constitutes virtually all of the H2A molecules. It has N-terminal and C-terminal region with a central globular domain. The C-terminal region has sites for modification including acetylation, biotinylation and phosphorylation.
Immunogen: synthetic phosphorylated peptide corresponding to the C-terminus (amino acids 134-142) of human histone H2AX (pSer139).
Immunogen: The synthetic peptide sequence is conjugated to KLH. The immunogen sequence is highly conserved (single amino acid substitution) in mouse histone H2AX.
