| Application: | Automated pilot scale purification of synthetic oligonucleotides; AKTAexplorer Summary This application note describes a convenient and simple protocol for the purification of synthetic phosporothioate oligonucleotides, using ion exchange chromatography on an automated AKTAexplorer 100 system. These data show that AKTAexplorer 100, controlled by UNICORN, is a convenient way of obtaining reliable and reproducible results, which can also be scaled-up. |

| General description: | RESOURCE™ Q columns are prepacked with SOURCE™ 15Q, a strong anion exchanger for intermediate lab-scale purification and large-scale polishing of biomolecules. The small (15 μm) monodisperse bead give high-resolution purification at high flow rates. In addition, hydrophilization of the beads minimizes nonspecific adsorption and allows high recovery of purified sample. The material of the column body is PEEK (polyetheretherketone). SOURCE™ 30Q and 30S media with a 30 μm bead size are designed for intermediate purification and large-scale polishing and allow higher flow rates. |
