β-Glucuronidase is routinely used to hydrolyze glucuronide-conjugates of drug metabolites in order to optimize sample preparation prior to LC-MS, GC and ELISA analysis.
LC-MS analysis of codeine-glucuronide enzymatic hydrolysis with six different Sigma-produced β-glucuronidase preparations. The limpet-derived enzyme G8132 performed best with this substrate, with reaction completion within 3–4 hours at 60 °C at 1 unit per mL. Comparative data above was performed at 60 °C at 10 unit per mL.
LC-MS analysis of hydromorphone-glucuronide enzymatic hydrolysis with six different Sigma-produced β-glucuronidase preparations. Molluscan enzymes performed similarly. The reaction was complete within 3–4 hours at 60 °C at 1 unit per mL. The E. coli enzymes exhibited low specificity for this substrate.
Enzymatic hydrolysis rates of six glucuronide substrates at 1,000 ng/mL in synthetic urine using heavy-labeled internal standards at 250 ng/ml. Hydrolysis was performed at 10 units/mL α-glucuronidase in acetate buffer at pH 5.0.

Application: The enzyme from patella vulgata is reported to be more effective in hydrolyzing opioid-glucuronides than the Helix pomatia, bovine liver and E. coli enzymes
Application: β-Glucuronidase Type L-II has been shown to be a superior enzyme for the hydrolysis of drug-glucuronides from urine.
Biochem/physiol Actions: β-glucuronidase (β-GIc) is an exoglycosidase that catalyzes the breakdown of complex carbohydrates. In humans it converts conjugated bilirubin into the unconjugated form, making bilirubin suitable for reabsorption. Morphine 3-β-D glucuronide was converted to morphine using ß-glucuronidase obtained from Patella vulgate.
Other Notes: One Sigma or modified Fishman unit will liberate 1.0 μg of phenolphthalein from phenolphthalein glucuronide per hr at 37 °C at pH 3.8 (30 min assay).
