Immunofluorescence
Localization of FMR1 in cytoplasmic stress like granules in NIH-3T3 cells. Cells were heat shocked for 30 min. at 43 ºC, fixed and stained with 2 μg/mL Rabbit Anti-FMR1 (C-terminal) (Cat. No. F4055). A. 100× magnification; B. 40× magnification.
Enhanced Validation-By Functional Assay Immunofluorescence. Antibody recognition: Anti-FMR1 (C-terminal) antibody produced in Rabbit (Cat. No. F4055) is confirmed by immunofluorescence in the NIH3T3 cell sample that has been treated with heat shock (Picture B) while no spefic staining is detected in the untreated sample-NIH3T3 (Picture A). Cells were counterstained with DAPI (blue) to stain nuclei. Antibody dilution 5 μg/mL.
Immunoprecipitation Rabbit Anti-FMR1 (C-terminal) (Cat. No. F4055) was used to immunoprecipitate human FMR1 from HEK-293T cell lysates. Lanes 1. 10 μg antibody. Detection antibody: Rabbit Anti-FMR1 (N-terminal) (Cat. No. F3930) 2-4. 10 μg, 5 μg and 3 μg antibody, respectively. Detection antibody FMR1 (C-terminal) (Cat. No. F4055) 5. Negative control – without cell lysate 6. Negative control – without IP antibody 7. Positive control – direct immunoblotting of HEK-293T cell extract
Immunoblotting
Lysates from various cell lines were separated on SDS-PAGE and probed with 2 μg/mL Rabbit Anti-FMR1 (C-terminal) (Cat. No. F4055). The antibodies were developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Lysates from the following cell lines were loaded:
Lanes: 1. HEK-293T; 2. HeLa; 3. A431; 4. RINSH; 5. RAT1; 6. PC12; 7. C2.

General description: FMR1 is a RNA binding protein expressed mainly in brain, neurons, placenta, testes and lymphocytes. Defect in FMR1 can lead to Fragile X Mental Retardation Syndrome due to lack of expression of FMR1 or expression of a mutant protein that cannot bind RNA. Anti-FMR1 (C-terminal) antibody can be used in immunofluorescence staining. Rabbit anti-FMR1 (C-terminal) antibody reacts specifically with FMR1.
General description: The FMR1 protein (fragile X mental retardation 1 protein) can bind to RNA. It contains two heterogeneous nuclear ribonucleoprotein K homology (KH) domains and one RGG box. Two proteins named FXR1 and FXR2 interact with FMR1. The protein is highly expressed in brain and testis.
Immunogen: synthetic peptide corresponding to amino acids 606-623 of human FMR1, conjugated to KLH. The corresponding sequence is highly conserved (1 amino acid substitution) in rat and mouse.
Physical form: Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
