Immunoperoxidase staining (PAP) of a fibrillary differentiated astrocytoma (WHO grade I, SAMS grade 1) using Rabbit Anti-Fibroblast Growth Factor-Basic (Cat. No. F3393). High bFGF expression is shown within tumor cells as well as within endothelial cells of preexistent capillary blood vessels. The tissue sample was paraffin-embedded and 4% Tris-buffered formalin-fixed without an enzymatic or antigen demasking pre-processing.

Immunoperoxidase staining (PAP) of a freshly explanted gliobastoma multiforme (WHO grade IV, SAMS grade 4) in culture (passage # 3) using Rabbit Anti-Fibroblast Growth Factor-Basic (Cat. No. F3393). Intracytoplasmatically, homogeneous, high bFGF expression is shown within highly polymorphic tumor cells. The tumor cells on glass coverslips were methanol/acetone (100%, ice-cold, 1:1) fixed without any enzymatic or antigen-demasking pre-processing.

Analysis Note: Bioactivity: Anti-Fibroblast Growth Factor-Basic (1-24) caused a dose-dependent decrease in the mitogenic activity of FGF-Basic such that a final dilution of 1:50 neutralized the bioactivity of 1 unit of FGF Basic.

Biochem/physiol Actions: Fibroblast Growth Factor Basic (also known as bFGF) is a pleotropic cytokine that that is a potent mitogenic factor. On binding to its cognate receptor tyrosine kinase, FGFR, bFGF is capable of activation of pathways such as MAPK and PI3K and produce cellular effects in varied cell types. Fibroblast Growth Factor-basic is reported to induce proliferation of fibroblasts, differentiation, wound healing and regeneration, limb formation, tissue remodeling and angiogenesis.

General description: Fibroblast growth factor-basic (FGFb), also known as NUDT6 (nudix (nucleoside diphosphate linked moiety X)-type motif 6) is encoded by the gene mapped to human chromosome 4q28.1. The FGFb gene spanning 34271 bp on genomic DNA contains 16 exons and six introns. FGFb RNA transcripts are expressed in various human tissues including liver, thymus, spleen, peripheral blood leukocytes, heart skeletal muscle, testis, colon and kidney.
Immunogen: synthetic peptide sequence corresponding to bovine FGF2 (1-24).
Physical form: Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline.
