Enhanced Validation-By Recombinant Expression Antibody recognition: Monoclonal Anti-c-Myc-FITC antibody produced in Mouse, Clone: 9E10 (Cat. No. F2047) is confirmed by immunofluorescence in the cell sample that has been altered to overexpress the target protein-HEK-293T over expressing c-Myc (Picture A) while no specific staining is detected in the wild type sample-HEK-293T (Picture C). Pictures B and D: cells were counterstained with DAPI (blue) to stain nuclei. Antibody dilution: 2.5 μg/mL.

Biochem/physiol Actions: c-Myc plays a vital role in regulation of cell growth, proliferation, differentiation and apoptosis. It is implicated in DNA replication, RNA splicing or transcription. Deregulation in the gene is associated with the development of various types of human cancers. An epitope located within amino acids 410-419, containing the sequence EQKLISEEDL of human c-Myc has been widely used as a tag in many expression vectors, enabling the expression of proteins as c-Myc tag fusion proteins. Epitope tags provide a method to localize gene products in a variety of cell types, to study the topology of proteins and protein complexes, and to identify associated proteins. In addition, it allows characterization of newly identified, low abundance or poorly immunogenic proteins when protein specific antibodies are not available.

General description: Monoclonal Anti-c-Myc (mouse IgG1 isotype) is derived from the 9E10 hybridoma, produced by fusion of mouse myeloma cells and splenocytes from BALB/c mice. The antibody is isolated from ascites fluid and conjugated to fluorescein isothiocyanate isomer I (FITC). Monoclonal Anti-c-Myc, FITC conjugate recognizes the c-Myc tag sequence on c-Myc tagged fusion proteins when expressed N- or C-terminal to the fusion protein. c-Myc is a proto-oncogene localized on long arm of human chromosome 8. The encoded protein contains an N-terminal transactivation domain.
Immunogen: synthetic peptide of the human p62c-Myc protein.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.
Preparation Note: For continuous use, store at 2-8 °C for up to one month.For extended storage, freeze in working aliquots.Repeated freezing and thawing is not recommended.Storage in “frost-free” freezers is not recommended. Ifslight turbidity occurs upon prolonged storage, clarifythe solution by centrifugation before use. Workingdilution samples should be discarded if not used within12 hours.
