Dual Labeling with CellVue® Claret and DAPI. Logarithmically growing U937 cells were labeled with CellVue Claret1,2 (final concentration: 107 cells per mL; 12.5 μM dye), cultured for 24 hours at 37 °C., fixed with 2% methanol-free formaldehyde in PBS, and counterstained ProLong Gold anti-fade reagent with DAPI. A Leica TCS SP-2 confocal microscope was used to image CellVue Claret (red pseudocolor; ex. 633 nm; detection: 410–499 nm) and DAPI (blue pseudocolor; ex. 405 nm; detection 652–800 nm) in a plane bisecting the nuclear DAPI signal. Membrane trafficking during the 37 °C incubation resulted in labeling of intracellular as well as plasma membranes. However, in contrast to covalent protein labeling with CFSE (not shown), CellVue Claret labeling did not penetrate the nuclear envelope, as evidenced by lack of co-localization between red and blue signals. Application: CellVue Claret kit fluoresces in the far red and may be used for general cell membrane labeling including in vitro cell labeling, in vitro lymphocyte proliferation studies, in vivo cytotoxicity assays, and also as a second color for monitoring cell-cell membrane transfer.
Biochem/physiol Actions: The CellVue Claret Fluorescent Cell Linker kit uses a proprietary membrane labeling technology to stably incorporate a far red fluorescent dye with long aliphatic tails (CellVue Claret) into lipid regions of the cell membrane.

Packaging: When using a 2 mL final staining volume containing a final concentration of 2 x 10-6 M of PKH26 dye:
• the MINCLARET kit provides enough dye for up to 25 samples of 20 million cells each and enough diluent for up to 5 samples of 20 million cells each;
• the MIDCLARET kit provides enough dye for up to 50 samples of 20 million cells each and enough diluent for up to 30 samples of 20 million cells each.
Users must determine the optimal dye and cell concentrations from their cell type(s) and experimental purposes.
