Dual Labeling with CellVue® Claret and DAPI. Logarithmically growing U937 cells were labeled with CellVue Claret1,2 (final concentration: 107 cells per mL; 12.5 μM dye), cultured for 24 hours at 37 °C., fixed with 2% methanol-free formaldehyde in PBS, and counterstained ProLong Gold anti-fade reagent with DAPI. A Leica TCS SP-2 confocal microscope was used to image CellVue Claret (red pseudocolor; ex. 633 nm; detection: 410–499 nm) and DAPI (blue pseudocolor; ex. 405 nm; detection 652–800 nm) in a plane bisecting the nuclear DAPI signal. Membrane trafficking during the 37 °C incubation resulted in labeling of intracellular as well as plasma membranes. However, in contrast to covalent protein labeling with CFSE (not shown), CellVue Claret labeling did not penetrate the nuclear envelope, as evidenced by lack of co-localization between red and blue signals.
