Whole cell lysates were separated on SDS-PAGE, blotted with Anti-CENP-A (Cat. No. C6867) and developed using Goat Anti-Rabbit IgG-Peroxidase (Cat. No. A0545) and chemiluminescent substrate. Antibody dilution 1:2,000. (Lane 1) HEK293 cells (untransfected); (Lane 2, 3) HEK293 cells transfected with human CENP-A; (Lane 3) Inhibition with CENP-A immunizing peptide (20 μg/mL).
Enhanced Validation-By Recombinant Expression Antibody recognition: Anti-CENP-A antibody produced in Rabbit (Cat. No. C6867) is confirmed by western blot in the cell sample that has been altered to overexpress the target protein-HEK-293T over expressing CENP-A (Lane 1) while no staining is detected in the wild type sample-HEK-293T (Lane 2). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Antibody dilution: 1:2,000.

Biochem/physiol Actions: The location of the centromere on the chromosome is dependent on the histone H3 variant centromere protein A (CENP-A). The N-terminal tail of CENP-A is highly different from other H3 variants. Serine 7 of (CENP-A) plays an important role in mitosis-specific phosphorylation. It also plays a key role in kinetochore assembly. Alteration in CENP-A causes various human cancers. CENP-A may play an important role in epithelial ovarian cancer (EOC) and may serve as prognostic marker. It acts as a potential target for gene therapy in the treatment of EOC. The NH2 terminus of human CENP-A is essential for mitotic progression and necessary for the platform function of CENP-A centromeric chromatin in the assembly as well as maintenance of active kinetochores.

General description: CENP-A is a 17 kDa histone H3- like protein.
General description: Centromere protein A is a protein encoded by the CENPA gene in humans. It is referred as CenH3 and CENP-A. It is a histone H3 variant that has centromere-specific nucleosomal property and is located at neocentromere found on a chromosome 10. CENP-A- binding clusters is associated with nucleosomal blocks within the neocentromeric chromatin.
Immunogen: synthetic peptide corresponding to amino acids 32-49 located near the N-terminus of human CENP-A, conjugated to KLH.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
