Application: TR146 cell line human has been used:
• in cytotoxicity studies with an antibacterial compound, metronidazole
• in viability and metabolic studies post blue laser light exposure,
• as an oral epithelium-resembling cell line model for in vitro C. albicans infection studies
• in barrier function studies using transepithelial electrical resistance (TEER)
• for the transport studies with C-reactive protein (CRP)
Cell Line Description: TR146 was derived from the neck node of a 67 year-old female (primary tumour was sited in buccal mucosa). The patient had had previous radiotherapy (6,000 rads) and neck dissection. TR146 is tumourigenic in female (nu/nu) mice leading to the production of rapidly progressing tumours. Histology: well-differentiated keratinizing squamous cell carcinoma.

Culture Medium: HAMS F12 + 2mM Glutamine + 10% Foetal Bovine Serum (FBS)

General description: The TR146 cells are appropriate for drug transport studies and they mimic normal human buccal epithelium. It serves as an oral mucosa model for barrier property studies with various molecules including tight junction proteins, aquaporins and epithelial-mesenchymal transition (EMT) markers.

Subculture Routine: Split sub-confluent cultures (70-80%) 1:4 to 1:10 seeding at approximately 3 x 104 cells/cm2 using 0.05% trypsin or trypsin/EDTA; 5% CO2; 37°C. Saturation density of 5 x 104 cells/cm2.
