Application: Basic fibroblast growth factor has been used as one of the components of EBM-B (endothelial Basal Medium-2 media, heat-inactivated fetal bovine serum, Glutamine-penicillin-streptomycin sulfate and basic fibroblast growth factor) media.
General description: Bovine bFGF has 146 amino acids. bFGF shares 55% amino acid homology with a second endothelial cell mitogen, acidic FGF (aFGF) (also known as endothelial cell growth factor, ECGF). The amino acid sequence for human bFGF is very similar to the sequence of the bovine protein. The proteins differ only by two amino acids. Both the human and bovine proteins appear to be synthesized initially with an amino-terminal extension of nine residues. bFGF is a potent mitogen for a wide variety of mesoderm- and neuroectoderm- derived cells including fibroblasts, vascular and corneal endothelial cells, myoblasts, chondrocytes, osteoblasts, smooth muscle cells, and glial cells. All cell types that respond to FGFs bear specific FGF cell surface receptors. In addition, bFGF induces blood vessel growth and wound healing in vivo. Other in vivo effects of bFGF are mesodermal induction in early embryos and limb and lens regeneration.
Other Notes: Recombinant Basic Fibroblast Growth Factor, human (hbFGF), is produced in E. coli and purified by standard chromatographic techniques.
Specific activity/EC50:<1 ng/ml, at least the same specific activity (EC50) compared to the indicated standard is guaranteed.
For life science research only. Not for use in diagnostic procedures.
Contents
Lyophilizate, before lyophilization filtered through a 0.2 μm pore size membrane
Preparation Note: Working concentration: 0.5 to 10 ng/ml
For use in serum-free cell culture.
Working solution: Reconstitute the lyophilizate in sterile PBS (final concentration 100 μg/ml). Dilute further with PBS or medium containing 1 mg/ml BSA or 1 to 10% serum
Storage conditions (working solution): -15 to -25 °C
Reconstituted solution can be stored in aliquots at -15 to -25 °C.
Stable in culture at least for 3 days.
Note: Avoid repeated freezing and thawing.
Quality: Endotoxin level: <0.1 EU/μg (LAL-test)
Note: 1 EU corresponds to 0.1 ng
Sequence: Chain Length 155 AA
Sequence: The primary structure of recombinant human bFGF (one polypeptide chain, 155 amino acids) is the biologically active precursor of natural human bFGF (one polypeptide chain, 146 amino acids), with the 9 additional amino acids located at the amino-terminus.
Specificity: Human bFGF is effective on mouse, bovine, and human cells.
Unit Definition: EC50 definition: The concentration of hbFGF that is required to support half-maximal stimulation of cell proliferation (MTT cleavage) with Balb/c 3T3 (NR6) cells.
