Titan One Tube RT-PCR System
Amplification of various RT-PCR products with the Titan One Tube RT-PCR System. RT-PCR was performed with cDNA synthesis at +50 °C and 35 PCR cycles. Aliquots of 15 μl were analyzed on a 1% agarose gel. cDNA synthesis can also be performed at +60 °C to help eliminate RNA secondary structure, if necessary. Lanes 1 + 14: DNA Molecular Weight Marker VII 2 + 13: DNA Molecular Weight Marker VI 3: 980-bp G3PDH (10 ng total human liver RNA) 4: 324-bp Β-actin (1 ng total human liver RNA) 5: 530-bp MCAD (10 ng total human liver RNA) 6 + 7: 1.9- and 4-kb dystrophin (10 ng total human skeletal muscle RNA) 8 + 9: 1-kb and 419-bp PSMA (100 ng total RNA from cell line LNCaP) 10: 145-bp HCV (approximately 5,000 copies) 11: 391-bp Interleukin-1Β/500 ng total human blood RNA 12: 192-bp M-CSF (100 ng total human liver RNA)

| General description: | The Titan™ One Tube RT-PCR System uses Reverse Transcriptase (RT) Avian Myeloblastosis Virus (AMV) for first strand cDNA synthesis and the Expand™ High Fidelity enzyme blend consisting of Taq DNA Polymerase and a polymerase with a proofreading activity for amplification of cDNA by polymerase chain reaction (PCR). The proofreading activity, leads to an increased PCR fidelity. The Titan One Tube RT-PCR System: • Reduces the error rate in PCR due to the proofreading capability. A threefold higher fidelity is obtained in comparision to Taq DNA polymerase. • Delivers increased sensitivity due to the high efficiency of all three enzymes. • Is faster than conventional two-step RT-PCR. |
