Test principle of the Isotype characterization with the IsoStrip Mouse Monoclonal Antibody Isotyping Kit
In the figure, mouse IgG1, kappa is shown. Cutting off the black area at the bottom of the strip is recommended for a permanent record. Procedure: 1. An antibody-containing sample is pipetted into a development tube, and agitated so that the colored latex beads are completely resuspended. 2. An isotyping strip is placed in the tube. 3. Within 1 - 5 minutes, a blue band will appear in either the kappa or lambda section of the strip, as well as in one of the class or subclass sections, indicating the class or subclass and light-chain composition of the monoclonal antibody. These blue bands will intensify as the sample moves up the strip. The positive control bands on each side of the isotyping strip should also appear, indicating the antibody-coated latex beads are functional and have traveled up the strip. After 5 minutes, the results can be interpreted.

Preparation Note: Working solution: Dilute Sample• Remove the desired number of isotyping strips from the canister. Remove the caps from an equal number of development tubes.
The tubes may be labeled with a pencil or felt-tipped lab marker for easy identification.
• Dilute a sample containing the mouse monoclonal antibody in 1% BSA/ phosphate-buffered saline (PBS), pH 7.2 to 7.6.
Culture supernatant samples should be diluted 1:10 to 1:100. Ascites samples should be diluted 1:20,000. These are recommended dilutions and may vary depending on the concentration of antibody in your sample. In our experience, a monoclonal antibody
concentration of 0.1 to 1 g/ml of diluted sample gives the best results. 150 ml of this diluted sample will be added to the development tube.
