BAKERBOND™ Standard Analytical Columns

J.T. Baker® Ion Exchange Columns

Wide-pore bonded phase packings used in these analytical columns are based on spherical 5µm silica gel with a mean pore diameter of 300Å. To this base are bonded a variety of functionalities to accomplish separations of a wide range of biomolecules.

These packings are chemically stable over the pH range 2 to 8.5 and to such cleaning and antimicrobial agents as sodium azide, dilute trifluoroacetic acid and acetic acid. They are also unharmed by organic solvents such as methanol and acetonitrile and by concentrated aqueous solutions of salts and urea.

Wide-pore packings are available in the following chemistries:

  • Carboxy-Sulfon (COOH and SO3H), a unique weak and strong cation exchanger also capable of chromatofocusing with simple buffers. Working pH range 2.0 to 10.0.
  • DEAM (diethylaminomethyl), a polyelectrolyte weak anion exchanger containing tertiary amines, similar to the commonly used DEAE matrix. Excellent resolution and capacity even at higher flow rates. At certain pH values, WP DEAM requires a higher salt concentration than DEAE-type matrices for protein elution.
  • QUAT is a polyelectrolyte strong anion exchanger containing quaternary amines. Can be used in separations requiring broad pH ranges up to pH 10. Excellent resolution for a wide variety of proteins.

The standard 4.6mm x 25cm column configuration provides the highest resolution and a typical operating back pressure of 1000 to 1500 psi at a flow rate of 1.0mL/min using aqueous mobile phase.

BAKERBOND™ Scout Columns

Scout 4.6mm x 5cm columns offer high resolution in a shorter time period. Back pressure is typically 200 to 400 psi at a flow rate of 1.0mL/min in an aqueous mobile phase. Low back pressure permits dramatic reductions in equilibrium time between column runs and increases the life span of the column and HPLC hardware.
